Abstract

Rimantadine hydrochloride (rimantadine) and amantadine hydrochloride (amantadine) are synthetic antiviral drugs,inhibitors of M2 membrane protein ion channel of the influenza A virus. Both amantadine and rimantadine are slowlyabsorbed after the oral administration and induce significant gastrointestinal adverse effects. In order to reduce the side effects and to improve pharmacokinetics of the studied drugs, cyclodextrin complexation has been proposed. This studyis dedicated to characterization, by means of infrared spectrometry, of amantadine and rimantadine products with βcyclodextrin (βCD) and hydroxypropyl-β-cyclodextrin (HPβCD) at the presence of a hydrosoluble polymer - polyvinylpyrrolidone (PVP). The corresponding ternary products were obtained using two main methods: kneading and coevaporation. In the result of this study we concluded that, as well as in the spectra of binary products studied before, the stretching vibrations of cyclodextrin glycosidic bonds and hydroxyl groups have significantly changed their frequencies in the spectra of AM-βCD-PVP and RIM-βCD-PVP products. At the same time these vibrations retained their frequencies unchanged in the spectra of AM-HPβCD-PVP and RIM-HPβCD-PVP products. Supposedly, the inclusion complexformation took place in the case of βCD-containing products, while the HPβCD-containing products remained uncomplexed.
