    SINTEZA ȘI CERCETAREA PROPRIETĂȚILOR FARMACOFORE ALE UNOR N-(DIMETILFENIL) HIDRAZINCARBOTIOAMIDE

    The present study was focused on the synthesis of some N-(dimethylphenyl)hydrazine carbothioamides 1-4, that contain the following N-substituents: 2,4-dimethylphenyl; 2,5-dimethylphenyl; 2,6-dimethylphenyl; 3,4-dimethylphenyl, to increase lipophilicity and N-(dimethylphenyl)-2-(pyridin-2-ylmethylidene)hydrazinecarbothioamides 5-8, analogous of Triapine. The structural formula of the compounds was characterized by means of spectroscopy: FT-IR, 1H-, and 13CRMN, and the molecular structure, for the first time, by means of X-ray diffraction. The study of antioxidant activity has shown that all compounds 1-8 are powerful antioxidants. N-(dimethylphenyl)-2-(pyridin2-ylmethylidene)hydrazinecarbothioamides 5-8 were tested as inhibitors of MCF-7 (breast cancer) cell proliferation. It was found that all the compounds exhibit activity comparable to that of Doxorubicin, among them the compound N-(2,5-dimethylphenyl)-2-(pyridin-2-ylmethylidene)hydrazinecarbothioamide 6, with IC50=0.8 μM/L, demonstrated the highest activity.

    SINTEZA COMBINAȚIILOR COORDINATIVE NOI ALE UNOR METALE 3D ÎN BAZA N(4)-(ACETAT DE ETIL) TIOSEMICARBAZONELOR 2-FORMIL ȘI 2-ACETIL PIRIDINEI. PROPRIETĂȚI ANTIOXIDANTE

    The given work includes the description of methods for the synthesis of new coordinative combinations of Cu(II), Mn(II), Fe(III) Co(III), Ni(II) and Zn(II) with N(4)-(ethyl acetate) thiosemicarbazones of 2-formylpyridine and 2-acetylpyridine. The characterization of the newly synthesized compounds was carried out using 1H NMR, 13C NMR, FTIR spectroscopy, conductivity, and elemental analysis. In addition, the structures of N(4)-(ethyl acetate) thiosemicarbazide and N(4)-(ethyl acetate) thiosemicarbazone 2-acetylpyridine (HL2) were determined using single crystal X-ray diffraction. The antioxidant activity of coordinative combinations was studied and compared with that of free ligands. The most pronounced antioxidant properties are possessed by [Ni(L2)Cl], whose value of IC50=15,05 μM is three times higher than the IC50=46,76 μM of the non-coordinating ligand and about 2,5 times higher than the reference substances Rutin and Trolox.

    SYNTHESIS, CHARACTERIZATION, ANTIOXIDANT ACTIVITY EVALUATION OF 3d METALS COMPLEXES WITH N(4)-((3)-ETHYL BENZOATE) THIOSEMICARBAZONES OF 2-FORMYL(2-ACETYL, 2-BENZOIL)PYRIDINE

    The paper presents the synthesis of new coordination compounds of Cu(II), Mn(II), Fe(III) Co(III), Ni(II) and Zn(II) with N(4)-((3)-ethyl benzoate)thiosemicarbazones of 2-formyl pyridine derivatives obtained through the condensation of N(4)-((m)-ethyl benzoate) thiosemicarbazide with 2-pyridinecarboxaldehyde, 2-acetylpyridine and 2-benzoylpyridine. The characterization of new compounds was done by 1H NMR, 13C NMR, IR spectroscopy, conductivity and elemental analysis. In addition, the structures of the ligands (HL1,3) were determined by single-crystal X-ray diffraction. The effect of these complexes on antioxidant activity were studied and compared with those of free ligands. The best result was found to be on coordination compound [Zn(L1)Cl], with IC50 = 11.45 μM, it is better than the free ligand HL1, IC50 = 13.15 μM and is better than the substance Trolox, IC50 = 15.86 μM (it is a pronounced antioxidant taken as a control substance).

    SINTEZA, STRUCTURA ȘI PROPRIETĂȚILE COMPUȘILOR COORDI NATIVI AI UNOR METALE 3 d CU 4 - (DIMETILFENIL) - TIOSEMICARBAZONE ALE 2 - HIDROXI - 3 - METOXIBENZALDEHIDEI

    A u fost sintetizate trei 4 - (dimetilfenil) - tiosemicarbazone (H 2 L 1 - 3 ) ale 2 - hidroxi - 3 - metoxibenzaldehidei şi 47 compuși coordinativi ai cuprului, cobaltului, nichelului și fierului cu azometine H 2 L 1 - 3 și diferite amine (A) sau sulfanilamide (Sf), pentru care în baza datelor analizei elementale a fost stabilită compozi ția: Cu(HL 1 - 2 )X·nH 2 O (X = Cl - , Br - , ClO 4 - , NO 3 - , n = 0, 4), Co(HL 1 - 2 ) 2 X (X = Cl - , NO 3 - ), Cu(L 1 - 3 )A ( A = H 2 O; Im; 2,2 ’ - Bipy; o - Phen; 3,5 - Br 2 py; Sf 1 - 4 ), Fe(HL 1 ) 2 X·nH 2 O (X = Cl - , NO 3 - ; n = 0, 2), Ni(L 1 - 2 )H 2 O și Ni(H 2 L 1 )(L 1 )· C 2 H 5 OH. În compoziția complecșilor sintetizați tiosemicarbazonele H 2 L 1 - 3 se comportă ca liganzi mono - sau tridentați neutri, mono - sau dublu - deprotonați. Cercetarea magnetochimică a demonstrat că compușii coordinativi ai cuprului au structură mononucleară sau polinucleară. În cazul compuși lor de cobalt și nichel s - au stabilit proprietăți diamagnetice. Compuşii coordinativi sintetizaţi nu manifestă activitate antimicrobiană faţă de Eschirichia coli , dar manifestă activitate bacteriostatică şi bactericidă faţă de microorganismele gram - pozitiv e şi fungi în diapazonul concentrațiilor 0,00006 – 1,0 mg/mL . S - a stabilit că asupra activității antimicrobiene a compușilor influențează natura atomului central, tiosemicarbazonei, aminei, sulfanilamidei și a restului de acid și pentru complecșii cu compo ziție asemămătoare se schimbă conform șirurilor: Cu  Ni ˃˃ Fe  Co; H 2 L 3 ˃ H 2 L 2 ˃ H 2 L 1 ; NO 3 -  Br - ˃ ClO 4 - ˃ Cl - ; 2,2 ’ - Bpy ˃ 3,5 - Br 2 Py ˃ Im ˃ o - Phen; Sf 4  Sf 3  Sf 2  Sf 1 . În diapazonul concentrațiilor 10 - 5 - 10 - 7 mol/L complecșii manifestă activitate ant ioxidativă de 1,6 - 9 ori mai înaltă decât activitatea antioxidativă a Troloxului , utilizat în medicină în calitate de etalon la determinarea proprietăților antioxidative ale compușilor naturali și sintetici.
