The antiproliferative properties of these compounds towards cancer cell lines HeLa, RD and normal cell line MDCK have been investigated. The tested compounds demonstrated high antioxidant and antiproliferative, selective activities towards cancer cells. Este utilizată metoda de biotestare toxicologică a compuşilor coordinativi CMT-28 şi CMA-18 asupra ciliatelor Paramecium caudatum Ehrnbg,în concentraţii de 100, 10, 1, 0,1, 0,01 uM/L, comparativ cu prototipul. Metoda de biotestare ne permite să formulăm rapid concluzii în plan comparat, evitând riscul unei erori înalte.Proprietăţile compuşilor metaloorganici au fost cercetate prin metoda diluărilor în serie, în mediul nutritiv lichid (1 g soluţie nutritivă/1 l apă). În calitate de culturi de referinţăau fost folosite ciliatele Paramecium caudatm. Infuzoriile, ca obiecte de cercetare, sunt foarte comode, se reproduc destul de rapid, fiind cele mai convenabile test-obiecte pentru investigaţii în condiţii de laborator, în care factorul timp joacăun rol decesiv, inclusiv pentru elucidarea impactului unor factori abiotici (substanţelor metaloorganice) asupra procesului de reproducere asexuatăla Paramecium caudatum. Activitatea substanţelor a fost evaluată după caracterul toxicităţii preparatelor (LT50 şi LC50), iar activitatea ciliatelor a fost studiată după caracterul viabilităţii şi parametrii reproductivi (creşterea sau diminuarea efectivului numeric şi a ratei lor de reproducere). Toxicitatea acută(LT50 şi LC50) a compuşilor a fost cercetată conform recomandărilor metodice privind studierea toxicităţii generale a remediilor farmaceutice. LC50(concentraţia letală, 50%) – concentraţia care cauzează moartea a 50% din test-organisme într-o anumită perioadă de timp. LT50(timpul letal, 50%) – timpul în care concentraţia utilizată a compusului testat cauzează moartea a 50% din organismele testate.Item Combinații coordinative ale Cuprului(II) Cu ({(2 [(piridin-2-il)metiliden]hidrazincarbotioil} amino) acetat de etil cu proprietăți antioxidative(CEP USM, 2024) Bîrcă, Maria; Rusnac, Anna; Garbuz, Olga; Evtodienco, Carolina; Gulea, AurelianIn this work, ethyl 2-isothiocyanatoacetate and [(hydrazinecarbotyoyl)amino]ethyl acetate, which are precursors of thiosemicarbazones, were synthesized.Based on these compounds, the synthesis of two new thiosemicarbazones ethyl 2-(2-(pyridin-2-ylmethylene)hydrazinecarbothioamido)acetate (Hl1) and ethyl 2-(2-(1-(pyridin-2-yl)-ethylidene)hydrazinecarbothioamide)acetate (Hl2) with yields between 85-88%. The thiosemicarbazones obtained were confrmed using 1H and 13C-nMR and FTIR spectroscopy. For the frst time, 8 coordination compounds were obtained based on thiosemicarbazone (Hl1 - Hl2) and Cu(II) salts: [Cu(l1)Br], [Cu(l1)(H2O)Cl], [Cu(l1)nO3], [Cu(l1)(H2O)ClO4], [Cu(l2)Br], [Cu(l2)(H2O)Cl], [Cu(l2)nO3], [Cu(l2)(H2O) ClO 4]. The composition of the synthesized coordination compounds was determined by elemental metal analysis and IR spectroscopy.For the synthesized compounds, the antioxidant capacity was evaluated by means of the aBTs method. The asymmetric unit of the unit cell in the crystal structures of I and II contains a copper complex with bidentate amine and a ligand coordinated by the azomethine nitrogen atom, the deprotonated phenolic oxygen atom, and the sulfur atom in the thiol form. The coordination polyhedron of the copper atom in compounds I and II is a distorted tetragonal pyramid. Obtained coordination compounds I and II exhibit antimicrobial and antifungal activities and have minimum inhibitory concentration and bactericidal concentration values in a range of 1.5-500 µg/mL. In this process, free radicals are constantly generated. It is widely known that free radicals play a dual role in vivo as both beneficial and adverse compounds. In order to maintain the balance of the activity of the free radicals in living cells, the organism has evolved antioxidant defense systems, protecting against free radical damage. However, when our endogenous defense system has incomplete efficiency, or a rise in free radicals under special conditions like smoking, chemical air pollutants, radiation, and inflammation, the imbalance between free radicals and antioxidants results in oxidative stress, which is now believed to be one of the important factors in the occurrence of certain human diseases including atherosclerosis, rheumatoid arthritis, cancer, and neurodegenerative diseases. So, the search of possible antioxidants agents is of interest, because they protect the body against damage by harmful free radicals. For this purpose two copper(II) coordination compounds of 1-(morpholin-4-yl) propane-1,2-dione 4- allylthiosemicarbazone (HL) with 1,10-phenantroline (1,10-Phen) and 2,2’-bipyridyl (2,2’- Bpy) were synthesized. On the first stage of the experiment the ethanolic solution of Cu(NO3)2⸱3H2O reacts with 4-allylthiosemicarbazone HL and on the second stage the ethanolic solution of the prepared precursor Cu(L)NO3 reacts with bidentate amines forming crystalline coordination compounds. The composition of these compounds was determined using elemental analysis for copper: Cu(1,10-Phen)(L)NO3, Cu(2,2’-Bpy)(L)NO3. The molar conductivity values of the synthesized coordination compounds are in the range 82-98 Ω- 1∙cm2∙mol-1 that indicates that these complexes are 1:1 electrolytes. A standard ABTS•+ method has been used to determine the antioxidant properties of synthesized compounds and trolox was used as a reference. The pro-ligand HL is less active than trolox, its coordination to copper(II) atom decreases antioxidant activity. Doxorubicina (7-[4-amino-5-hidroxi-6-metiloxan-2-il]oxi-6,9,11-trihidroxi-9-(2-hidroxiacetil)4-metoxi-8,10-dihidro-7H-tetracen-5,12-dionă) a fost utilizată în calitate de compus anticancerigen de referință și Trolox (acid 6-hidroxi-2,5,7,8-tetramethilcroman-2-carboxilic) a fost utilizat ca compus antioxidant de referință. Compușii testați au demonstrat efect antioxidant înalt față de cationii radicali ABTS•+ în comparație cu interacțiunea acestor cationi cu Trolox. Compușii testați au manifestat toxicitate generală față de Daphnia magna după 24 de ore de expunere cu determinarea unui șir de descreștere. Experiențele clasice cu ciliatul standard Paramecium caudatum (Ehrenberg, 1833) demonstrează un efect stimulator neașteptat asupra viabilității acestora, crescând semnificativ rata lor de reproducere la concentrații micromolare ale sării de litiu în mediul nutritiv.Item Exploring the antioxidant activity of 1-(piperidin-1-yl) propane-1,2-dione 4-phenylthiosemicarbazone and 3d metal coordination compounds with this ligand(CEP USM, 2024) Graur, Ianina; Maxim, Mihail; Graur, Vasilii; Tsapkov, Victor; Gulea, Aurelian; Garbuz, OlgaThe new 1-(piperidin-1-yl)propane-1,2-dione 4-phenylthiosemicarbazone (HL) and its 7 coordination compounds [Cu(L)X] (X = Br- (I); Cl- (II); NO3 - (III); CH3COO- (IV); Cl2CHCOO- (V)), [Co(L)2]NO3 (VI), [Fe(L)2]NO3 (VII) have been synthesized and characterized by elemental analysis, FTIR, 1H, 13C NMR spectra and molar electric conductibility. For the obtained compounds it was studied the antioxidant activity towards ABTS•+ radical cation. Uncoordinated 4-phenylthiosemicarbazone HL is the most active one with IC50 14.1 μM. Its activity is higher than that of Trolox which is used in medicine as an antioxidant. Among all the synthesized coordination compounds, the iron(III) complex VII showed the greatest activity. The antioxidant activity of metal complexes is influenced by the anion and the nature of the central metal atom.Item IN VITRO ANTICANCER ACTIVITY OF CHLORO(N-PHENYL-N'-[(PYRIDIN-2- YL)METHYLIDENE]CARBAMOHYDRAZONOTHIOATO) (4-AMINOBENZENE-1-SULFONAMIDE)COPPER(CEP USM, 2018) Garbuz, OlgaThis study was aimed to evaluate the antiproliferative activity of the mixed-ligand complex (chloro(N-phenyl-N'- [(pyridin-2-yl)methylidene]carbamohydrazonothioato)(4-aminobenzene-1-sulfonamide)copper) on several cancer cells of lines. It was established, that the copper(II) mixed-ligand complex exhibits the highest anticancer activity against MeW-164, HeLa, BxPC-3 and RD cells of lines with IC50 values of 1.0±0.2, 0.4±0.04, 1.7±0.2, 1.3±0.3 µM, respectively. A comparative study between the tested compound and DOXO in regard to cancer lines has established that the tested copper(II) mixed-ligand complex exhibits stronger inhibitory activity on cancer cells proliferation than doxorubicin and cisplatin.Item IN VITRO ANTIPROLIFERATIVE ACTIVITY AND ANTIOXIDANT CAPACITY OF NEW ORGANOMETALLIC COORDINATION COMPOUNDS, RESULTS CORRELATION ANALYSIS(2016) Gulea, Aurelian; Gudumac, Valentin; Garbuz, Olga; Andronache, LiliaA fost examinată și testată capacitatea inhibitorilor organometalici activi CMJ-33, CMSA-4, de a induce apoptoza în celulele epiteliale de carcinom de col uterin linia HeLa. Viabilitatea celulelor canceroase a fost determinată prin testul Alamar Blue fiind folosit un indicator redox – resazurin. S-a depistat activitate antitumorală mai mare a substanțelor testate decât la cea luată în calitate de etalon: - doxorubicina (DOXO). În scopul de a detecta eventuala prezenţă a efectelor adverse concomitente legate cu proprietățile pro-oxidante ale compușilor coordinativi, a fost folosită metoda cantitativă ORAC (Oxygen Radical Absorption Capacity). Rezultatele au demonstrat că substanțele testate CMT-67 şi CMSA-4 posedă activitate antioxidantă mai mare decât Trolox și DOXO. A fost studiată sensibilitatea culturii de laborator P. caudatum la acţiunea substantelor coordinative și posibilitatea de adaptare a cesteia la diferite concentrații. Toxicitatea a fost evaluată prin testarea parametrilor reproductivi și compararea lor cu martorul. A fost calculată dinamica, statistic semnificativă, dintre procesul de adaptabilitate și toxicitatea ciliatelor Paramecium caudatum la diferite concentraţii. În final, au fost determinate concentraţiile optime ale unor substanţe şi ale celor care au o influență toxică asupra viabilității organismelor unicelulare
