    SYNTHESIS, CHARACTERIZATION, ANTIMICROBIAL AND ANTIPROLIFERATIVE ACTIVITY EVALUATION OF CU(II), CO(II), ZN(II), NI(II) AND PT(II) COMPLEXES WITH ISONIAZID-DERIVED COMPOUND

    Hydrazone complexes of Cu(II), Co(II), Zn(II), Ni(II) and Pt(II) with N-isonicotinoyl-N′-(3-metoxy-2 hydroxybenzaldehyde)-hydrazone (HL) were synthesized and characterized by different physico-chemical techniques including elemental and thermal analysis, magnetic susceptibility measurements, molar electric conductivity, as well as IR (infrared), 1H-NMR and 13C-NMR (hydrogen and carbon nuclear magnetic resonance, UV-Vis (ultraviolet-visible), FAB (fast atom bombardment), EPR (electron paramagnetic resonance), and mass spectroscopies. The crystal structure of ligand was determined by single crystal X-ray diffraction studies. Spectral data showed that hydrazone behaves as an ONO tridentate ligand through the azomethine nitrogen, phenolate and keto oxygen atoms. For the copper(II) complexes, metal-ligand bonding parameters were evaluated from the EPR spectra. These parameters indicate the presence of in-plane π bonding. In addition, the f values of complexes 1-4 indicate small distortion from planarity. The effect of these complexes on proliferation of human breast cancer (MCF-7 and SKBR-3), human melanoma (A375), lung adenocarcinoma cells (NCI-H1573) and their antibacterial activity against Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus and Candida albicans strains were studied and compared with those of free ligand. The ligand and complexes 1-3 showed significant antimicrobial activity against the Gram-positive bacteria Staphylococcus aureus and Candida albicans in comparison to the control drugs. The complexes 2-4 could be potential antitumor agents, leading to a significant improvement of the cytotoxic activity when compared with HL.

    SYNTHESIS AND CHARACTERIZATION OF NOVEL CU(II), PD(II) AND PT(II) COMPLEXES WITH 8-ETHYL-2-HYDROXYTRICYCLO(7.3.1.02,7)TRIDECAN-13-ONE- THIOSEMICARBAZONE: ANTIMICROBIAL AND IN VITRO ANTIPROLIFERATIVE ACTIVITY

    New Cu(II), Pd(II) and Pt(II) complexes, (Cu(L)(H2O)2(OAc)) (1), (Cu(HL)(H2O)2(SO4)) (2), (Cu(L)(H2O)2(NO3)) (3), (Cu(L)(H2O)2(ClO4)) (4), (Cu(L)2(H2O)2) (5), (Pd(L)(OAc))H2O (6), and (Pt(L)2) (7) were synthesized from 8-ethyl-2-hydroxytricyclo(7.3.1.02,7)tridecan-13-one thiosemicarbazone (HL). The ligand and its metal complexes were characterized by IR, 1H-NMR, 13C-NMR, UV-Vis, FAB, EPR, mass spectroscopy, elemental and thermal analysis, magnetic susceptibility measurements and molar electric conductivity. The free ligand and the metal complexes have been tested for their antimicrobial activity against E. coli, S. enteritidis, S. aureus, E. faecalis, C. albicans and cytotoxicity against the NCI-H1573 lung adenocarcinoma, SKBR-3 human breast, MCF-7 human breast, A375 human melanoma and HL-60 human promyelocytic leukemia cell lines. Copper complex 2 exhibited the best antiproliferative activities against MCF-7 human breast cancer cells. A significant inhibition of malignant HL-60 cell growth was observed for copper complex 2, palladium complex 6 and platinum complex 7, with IC50 values of 1.6 μM, 6.5 μM and 6.4 μM, respectively.

    ANTIMICROBIAL AND ANTIFUNGAL ACTIVITY OF CU(II) AND BI(III) COMPLEXES BASED ON AMINOPOLYCARBOXYLATE IONS AND 2-FORMYL AND 2-ACETYLPYRIDINE THIO- SEMICARBAZONES

    The biological properties of thiosemicarbazones are often related to metal coordination, which as result affects lipophilicity that controls the rate of entry into the cell. Metal complexes are generally more active than the free ligand, or the metal complex can be a vehicle for activation of the ligand as the cytotoxic agent. Among biologically ac- tive metal compounds, copper (II) complexes with 2-formyl and 2-acetylpyridine thio- semicarbazones proved to be among the most potent antiviral, antitumor and anti-inflam- matory agents. Bismuth compounds have been widely used in medicine for more than two centuries due to their high effectiveness and low toxicity. In the antimicrobial realm, ap- plications have been widespread, due to bismuth antiseptic, astringent, protective, ant- acid, antisecretory and local gastrointestinal properties.

    SYNTHESIS, CHARACTERIZATION AND TOXICITY EVALUATION OF CU(II), MN(II), CO(II), NI(II), PD(II) COMPLEXES WITH LIGAND DERIVED FROM HYDRAZINECARBOTHIOAMIDE

    New copper(II), manganese(II), cobalt(II), nickel(II) and palladium(II) complexes, [Cu(L)(X)(H2O)] [X= OAc- (1), Cl- (3), ClO4 - (4)], [M(L)2(H2O)2] [M= Cu2+(2), Mn2+(5)], [M(L)(OAc)(H2O)] [M= Co2+(6), Pd2+(8)], [Ni(L)(OAc)] (7) were synthesized with 2-(5H-dibenzo [a,d][7] annulen-5-ylacetyl)-N-(4-chlorophenyl) hydrazinecarbothioamide (HL). All the metal complexes have been characterized by different physico-chemical techniques like molar conductivity, magnetic susceptibility measurements, electronic and infrared spectral studies, elemental and thermal analysis. The toxicity of newly synthesized compounds was evaluated using the Daphnia magna bioassay.

    SINTEZA, STRUCTURA ȘI PROPRIETĂȚILE COMPUȘILOR COORDINATIVI AI CUPRULUI(II) CU DERIVAȚII TIOSEMICARBAZONEI 8 - FORMILCHINOLINEI

    În acest studiu este descrisă sinteza și analiza cristalografică a compusului coordinativ al cuprului(II) cu tiosemicar-bazona 8-formilchinolinei. În soluţii diluate la temperatura camerei, reactivii organici reacţionează cu ionii de cupru(II)formând compuşi de culoare galbenă, cu raportul de combinare metal:ligand = 1:1. Compusul coordinativ al cuprului(II) cu 4-feniltiosemicarbazona 8-formilchinolinei este caracterizat spectral printr-un maxim de absorbție localizat la at λ =390 nm.Curba de calibrare are un caracter liniar în domeniul de concentraţii ale cuprului 2,5∙10-6÷ 4∙10-5M. 4-fenil-tiosemicar bazona 8-formilchinolinei a fost propusă în calitate de reactiv pentru determinarea spectrofotometrică a cuprului. Metoda de determinare cantitativă a cuprului este binevenită în cazul când randamentul reacţiilor de formare a compuşilor coordinativi cu liganzi organici este mic. Compusul [CuLAc] manifestă activitate antibacteriană față de bacteriile Staphylococcus aureus(tulpina 209-P), Entrecoccus faecalisși Proteus vulgaris.

    SINTEZA ŞI STRUCTURA COMPUŞILOR COORDINATIVI AI CUPRULUI(II) CU TIOSEMICARBAZONA ALDEHIDEI 5-(M ETILENTRIMETILAMONIU)SALICILICE

    A fost sintetizată clorura tiosemicarbazonei aldehidei 5-(metilentrimetilamoniu)salicilice în formă de sare ((H2L)Cl•1.5H2O), iar la interacţiunea acesteia cu diferite săruri de cupru(II) în mediu neutru au fost obţinuţi patru compuşi cu formulele [Cu(HL)Cl]Cl•2.25H2O (1), Cu(HL)(Cl)(NO3)]•2H2O (2), [Cu(H2L)Cl]SO4•0.75H2O (3) şi [Cu(HL)Cl]ClO4•1.75H2O (4), ce au fost stabilite cu ajutorul razelor X. Trei compuşi ai cuprului sunt compuşi ionici, formaţi din cationul complex [Cu(HL)Cl]+ori [Cu(H2L)Cl]2+, anionii fiind Cl-, ClO4- şi SO42- respectiv, iar compusul ce conţine anionul NO3-este un complex molecular. Toţi compuşii conţin molecule de apă de solvatare. Liganzii (HL)0/(H2L)+în 1, 2, 4/3coordinează tridentat prin setul de atomi donori O,N,S în formă de ion bipolar, singur fiind electroneutru, însă conţinând două centre ionice (Zwitter-ion) ori în forma sa protonată, respectiv. Raportul molar de combinare Cu:(HL)0/(H2L)+în aceşti compuşi este 1:1. În rezultatul coordinării liganzii organici (HL)0/(H2L)+suferă schimbări conformaţionale, iar primii sunt supu şi monodeprotonării.
